The PathHunter mCNR2 eXpress kit provides an easy-to-use cell based assay to study drug candidates (small molecules or biologics) targeting GPCRs. This kit assesses ligand (e.g. CP 55,940) based activation of mCNR2 GPCR activity via detection of β-arrestin recruitment. The included cell line overexpresses ProLink-tagged mCNR2 and EA-tagged β-Arrestin-2. Activation of mCNR2 stimulates the recruitment of β-Arrestin-2 and produces EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. This detection kit is a robust, sensitive and easy-to-use assay to determine GPCR activity through detection of intracellular cAMP levels. The HitHunter cAMP assay is intended for use with cells expressing Gαs- or Gαi-coupled GPCRs. This product uses EFC technology, and contains ED-tagged cAMP, anti-cAMP antibody, and pure EA. Upon application to a cell which contains endogenous, untagged cAMP, the anti-cAMP antibody will be sequestered by binding to this untagged cAMP, allowing for EFC between the tagged cAMP and soluble EA. This will produce a proportional level of chemiluminescent signal. This kit format is intended for Biologic drugs with a protocol optimized for use in a 96-well plate. This kit may be used with Eurofins DiscoverX cAMP cell lines as well as cell lines from other sources. This detection kit contains reagents for assaying 2 plates (96-well).
